Is it possible to set up a wireless router as an access point to an existing wireless network without a cable?

I am using a belkin wireless router to interface with my ISP. I have a big house and cannot get the signal everywhere. I have a spare linksys wireless router that I want to use as an access point without actually plugging it into the belkin router. Can this work?

Yes, depending on the precise router type. You want to configure the spare router as a bridge using WDS; if the router doesn't support it you may be able to download firmware to do it (just look for your router type and WDS or wireless bridge).
